Discovery NotesPhage was extracted from the dark, moist, clumpy soil of the Erickson Courtyard on the campus of the University of Maryland Baltimore County, via direct plating method. Phage was purified via streaking and phage titer. A medium titer lysate (titer: 10^9) and high titer lysate (titer: 10^11) was generated and the DNA was isolated from the HTL. DNA concentration via nanodrap was 433.1 ng/uL. Restriction analysis was conducted and the DNA was cut by Cla1, EcoRI, and HindIII, and the genome size was estimated to be 79 kbp. QC gel showed that the DNA was good quality and possessed little smearing, and had the concentration of 125-250 ng/uL. PCR gel showed one product in the C2 lane.
